To do this, Dr. Agata Patriotis utilizes both genetically engineered mouse models (GEMMs) as well as mouse and human-derived organoids. Specifically, she is performing bulk and single-cell assays for transposase-accessible chromatin using sequencing (ATAC-sequencing) to profile the chromatin state in this model. Dr. Agata aims to study transcriptional and epigenetic mechanisms that drive tumor progression and metastasis in the KP model. Additionally, she is studying how mutations in the epigenetic regulators frequently mutated in PDAC influence tumors' epigenetic state and cellular plasticity.
